Human insulin

Insulin, a polypeptide hormone with a molecular weight of 5800 Da, is secreted by the beta cells of the islets of Langerhans from the pancreas. Insulin possesses a wide spectrum of biological actions. It stimulates cellular glucose uptake, glucose oxidation, glycogenesis, lipogenesis, proteogenesis and the formation of DNA and RNA. Insulin plays a key role in the regulation of plasma glucose levels (hepatic output inhibition, stimulation of peripheral glucose utilisation).

S-type ELISA has a very low cross-reactivity to proinsulin.

We tried a recovery test by adding proinsulin to assay samples. The assay results are shown below. It proved that proinsulin added to the samples only slightly increased the insulin assay value. This means we can measure insulin precisely in the presence of proinsulin.

As shown in the figures, very high correlation coefficients were obtained with both rat(R=0.9946) and mouse(R=0.9868) samples.

The slopes of the regression lines were 1.209 in rat and 1.198 in mouse. This means that type S kits gave about 20% lower assay values than ordinary T-type kits with both rat and mouse samples. This must be caused by improvement of specificity in S-type. Though we should be careful in thinking that the difference is directly related proinsulin, difference of 20% is a reasonable value of proinsulin ratio to IRI in human subjects.

Proinsulin Intact

Proinsulin (PI), a polypeptide of 9390 MW (86 amino acids) is synthetized in the ß cells of the Islets of Langerhans in the pancreas. The level of proinsulin in serum can be a reflection of marker cell status and a consequence of disfunction of PI processing and/or secretion. Proinsulin like material is increased in clinical conditions as insulinoma, familial hyperinsulinemia, non-insulin dependent diabetes mellitus.

Insulin is synthesized in the beta-cells of the islets of Langerhans as a precursor molecule, proinsulin. In the secretory granules of the beta-cells, proinsulin is cleaved into insulin and into a 31-amino?acid peptide, called the Connecting Peptide or C?Peptide. Insulin and C-Peptide are secreted in equimolar amounts. However, because of its longer half-life, the plasma concentration of C-peptide is higher than that of insulin.

The determination of plasma C-Peptide allows an assessment of the endogenous insulin production, even in the presence of exogenous insulin administration or in the presence of circulating anti?insulin antibodies

Clusterin

Adiponectin

Quantification de l'activité métabolique liée au glucose grâce à un petide marqué pour la détection des récepteurs Glut-1 : EGFP-GLUT1 ligand

Le transport du Glucose, médié par un ou plusieurs membre des transporteurs du glucose (GLUT) joue un rôle essentiel dans le métabolisme des cellules normales ou pathologiques. GLUT-1 est l'isoforme le plus largement exprimé des transporteurs du glucose connus et est le principal transporteur du glucose à travers la barrière hémato-encéphalique. Les forts niveaux d'expression de GLUT-1 dans les cellules tumorales sont associés aux mauvais pronostics d'évolution de différents types de cancers, dont les cancers du sein et de l'estomac (2). Des recherches récentes montrent que GLUT-1 effectue le transport à travers la barrière hémato-encéphalique de certaines molécules neuro-actives comme les peptides glycosylés. Il a été démontré que le peptide produit par AbCys : EGFP-GLUT-1 ligand se lie à la partie carboxy-terminale de la boucle extracellulaire de GLUT-1 (contrairement à la plupart des anticorps existants, qui reconnaissent la totalité de la molécule(3) Ce peptide permet de mesurer l'expression de GLUT-1 sur les membranes des cellules de mammifères par FACS ou marquage fluorescent. (4) et peut procurer des renseignements, en atnt que marqueur qualitatif et quantitatif sur base moléculaire de l'homéostasie normale ou pathologique du glucose, comme le diabète ou comme deux désordres génétiques , le syndrome Fanconi-Bickel ou le syndrome de la maladie De-Vivo(6) . La détermination de l'expression de GLUT-1 par la liaison du ligand est corrélée à l'augmentation de l'absorption du glucose observée dans les cellules malignes et les cellules au métabolisme actif. EGFP- GLUT-1 ligand a permis de mesurer l'expression de GLUT -1 pendant l'activation de lymphocytes T et a été déterminant pour l'identification d'une population de thymocytes immatures CD4+ CD8 + possèdant une forte activité métabolique. Human embryonic kidney 293T cells and human erythrocytes (RBC) were incubated for 30 min at 30°C with a 1:25 dilution of EGFP-GLUT1 ligand. Staining profiles were analyzed by FACS and histograms depicting staining with « mock » protein are indicated.
