Streptavidin-Biotin System for BrdU Staining
Introduction
In the past, cell-proliferation studies used radioactive thymidine as an incorporated label to give evidence of DNA replication. The BrdU staining kit uses a less hazardous technique by using bromodeoxyuridine (BrdU), a thymidine analog, in lieu of a radio-active reagent.(1) BrdU is incorporated into proliferating cells (S-phase) much in the same way as radioactive thymidine, but is then detected by a monoclonal anti-BrdU antibody, and revealed using a highly sensitive streptavidin-biotin staining system. BrdU Staining Kit uses a biotinylated monoclonal anti-BrdU, thus eliminating the need for a species-specific secondary antibody. As a result, BrdU labeling can be performed in rats and mice without problems of cross-reactivity or spurious staining.
Expected staining results
BrdU labeling by IHC is specifically exhibited in the nuclei of cells in BrdU-incorporated tissue sections. The use of the included positive control slides is recommended. The images below of BrdU labeling by IHC also serve as a reference.

The BrdU labeling reagent is supplied as a concentrated aqueous solution of 5-bromo-2'-deoxyuridine and 5-fluoro-2'-deoxyuridine (10:1).

Description: This monoclonal antibody reacts with Bromodeoxyuridine (BrdU) and cross reacts with iododeoxyuridine (IdU). ZBU30 will react with BrdU incorporated into cells of any species. Detection of incorporated BrdU is useful in identifying cells in the S phase of the cell cycle, which can be important in studying the roles of various factors in cell cycle regulation. The antibody is a useful cellular proliferation marker. Immunogen: BrdU-bovine globulin. Positive tissue: BrdU labeled tissue.
