Adiponectin

Adiponectin, also referred to as Acrp30, AdipoQ and GBP-28, is a recently discovered 244 aminoacid protein, the product of the apM1 gene, which is physiologically active and specifically and highly expressed in adipose cells.
Adiponectin has been shown to increase insulin sensitivity and decrease plasma glucose by increasing tissue fat oxidation. Of particular interest is that low adiponectin serum levels predict type 2 diabetes independent of other risk factors.
Adiponectin has the potential to become a clinically relevant parameter to be measured routinely in subjects at risk for type 2 diabetes, atherosclerosis and the metabolic syndrome

Insulin

Insulin, a polypeptide hormone with a molecular weight of 5800 Da, is secreted by the beta cells of the islets of Langerhans from the pancreas. Insulin possesses a wide spectrum of biological actions. It stimulates cellular glucose uptake, glucose oxidation, glycogenesis, lipogenesis, proteogenesis and the formation of DNA and RNA. Insulin plays a key role in the regulation of plasma glucose levels (hepatic output inhibition, stimulation of peripheral glucose utilisation).

Proinsulin Intact

Proinsulin (PI), a polypeptide of 9390 MW (86 amino acids) is synthetized in the ß cells of the Islets of Langerhans in the pancreas. C-Peptide

Insulin is synthesized in the beta?cells of the islets of Langerhans as a precursor molecule, proinsulin. In the secretory granules of the beta?cells, proinsulin is cleaved into insulin and into a 31-amino?acid peptide, called the Connecting Peptide or C?Peptide. Insulin and C-Peptide are secreted in equimolar amounts. However, because of its longer half?life, the plasma concentration of C?peptide is higher than that of insulin.

Leptin

Leptin is produced mainly in the adipose tissue, and is considered to play an important role in appetite control, fat metabolism and body weight regulation. . Clusterin

Clusterin (Apolippoprotein J; SP-40,40; TRPM-2; SGP-2; pADHC-9; CLJ; T64; GP III; XIP8) is a highly conserved disulfide-linked secreted heterodimeric glycoprotein of 75-80 kDa but truncated forms targeted to nucleus have also been identified. Recent study demonstrate, that serum Clusterin level increases significantly in diabetic type II patients and in patients with developing coronary heart disease, or myocardial infarction. These date raise the possibility that elevated Clusterin levels in serum may represent a strong indication of vascular damage.

EGFP-conjugated GLUT1 ligand

Description:

The only detection of extra facial domain of GLUT1 on mammalian cells.

Glucose transport, as mediated by the members of the closely related glucose transporters (GLUT), plays an essential role in normal and pathological cellular metabolism. GLUT1 is the most widely expressed isoform of the known glucose transporters and the primary glucose transporter across the blood-brain barrier (1). High levels of GLUT1 expression on tumors have been associated with worse prognosis in different types of cancers, including breast and gastric carcinoma (2,3). Also, recent investigations have shown that GLUT1 mediates the transport of some glycosylated neuroactive peptidic drugs designed to cross the blood-brain barrier (4).

The recent identification of the receptor binding domain of the human T cell leukemia virus (HTLV) envelope glycoprotein (HRBD) that is necessary and sufficient to bind GLUT1 (5,6) has allowed the development of an HRBD derived peptide coupled with EGFP (EGFP-GLUT1 ligand).

EGFP-GLUT1 ligand has been shown to bind the carboxy terminal extracellular loop of GLUT1 (7) and has allowed GLUT1 expression to be monitored at the plasma membrane of all mammalian cell lines by FACS analyses or fluorescence staining (8). EGFP-GLUT1 ligand has facilitated studies of GLUT1 expression during T cell activation (9) and has been critical for the successful identification of an immature CD4+CD8+ human thymocyte population with high metabolic activity (10). Assessment of GLUT1 expression via binding of EGFP-GLUT1 ligand correlates with increased glucose uptake, as observed in malignant cancer cells or metabolically active cells (see fig 3). EGFP-GLUT1 ligand can provide insights, as a qualitative and quantitative marker, into the molecular bases of both normal and disordered glucose homeostasis.

Human embryonic kidney 293T cells and human erythrocytes (RBC) were incubated for 30 min at 30°C with a 1:25 dilution of EGFP-GLUT1 ligand. Staining profiles were analyzed by FACS and histograms depicting staining with « mock » protein are indicated.
